James Matthews Duncan was born in Aberdeen and educated at Aberdeen Grammar School. He obtained an MA from Marischal College before studying medicine in Aberdeen and Edinburgh. He received his MD from Aberdeen in 1846 and became a Licentiate of the Royal College of Surgeons of Edinburgh in 1847. He was appointed junior assistant to Professor James Young Simpson and participated in the early chloroform experiments.
